Overview

The PaldiSpot test detects and quantifies Borrelia, Babesia, and Bartonella genospecies in a patient’s DNA. Working from our international world-class facilities, our scientists use a highly sensitive molecular biology technique known as quantitative polymerase chain reaction (qPCR) to analyse DNA extracted from a blood sample. This advanced method screens for the presence of 24 of the most common strains of Borrelia, Babesia, and Bartonella genospecies with exceptional precision.

For patients with a positive Lyme disease diagnosis within the last 6 months, the PaldiSpot test provides critical information that can inform the development of personalised treatment and therapy plans for Lyme disease and related co-infections — helping clinicians make more informed decisions and improving patient outcomes.

The PaldiSpot test is clinically validated and highly accurate. It is suitable for adults aged 18 and over. Results are processed and returned within 14 business days.

Purpose of The Test

The PaldiSpot test analyses DNA extracted from blood cells and serum to screen for 24 of the most common Borrelia, Babesia, and Bartonella genospecies.

During the testing process, the patient’s DNA is used in quantitative polymerase chain reaction (qPCR) — an advanced laboratory method that amplifies and measures specific DNA sequences in real time. Each sample is examined for the genetic signatures of 24 target strains.

To ensure the highest level of reliability, all reactions are performed in triplicate, with both positive and negative controls conducted for every strain tested.

The final PaldiSpot report indicates whether a specific strain is present in the patient’s cells or serum and includes a quantification parameter known as the Ct value (cycle threshold), which reflects the detected genetic load.

Scientific Background

The PaldiSpot test is based on quantitative polymerase chain reaction (qPCR) — a highly sensitive molecular biology technique that allows scientists to detect and measure even the smallest quantities of genetic material. qPCR works by amplifying specific DNA sequences in real time, which enables precise identification of microbial DNA in a patient’s sample.

In the PaldiSpot test, DNA is extracted from blood cells and serum, then analysed for 24 of the most clinically relevant Borrelia, Babesia, and Bartonella genospecies — pathogens commonly linked to Lyme disease and associated co-infections. Each reaction is performed in triplicate, with positive and negative controls included for every strain to ensure reproducibility and diagnostic accuracy.

The test measures amplification cycles to generate a Ct value (cycle threshold), which reflects the relative amount of microbial DNA present in the sample. This allows clinicians not only to confirm the presence of infection but also to estimate pathogen load, providing valuable insight into disease activity and progression.

When to Consider the Test

The PaldiSpot test is typically recommended for patients with a clinical diagnosis of Lyme disease or related co-infections within the last 6 months, particularly those presenting with compatible symptoms or a documented history of exposure, such as a tick bite.

Please note: To be eligible for the PaldiSpot test, you must have received a positive diagnostic test result for Lyme disease within the last six months.

It may also be used:

  • Alongside the PrimeSpot test, to provide a broader assessment of infections linked to Lyme disease and chronic fatigue syndromes.
  • Following inconclusive or negative standard serological tests, to provide additional molecular evidence.
  • For monitoring, to evaluate the effectiveness of treatment or track disease progression.

At this time, the PaldiSpot test is not designed for preventive screening in asymptomatic individuals or for general population testing.

Samples & Processing

Once the test is ordered, your Biocentaur approved clinician will be provided with a PaldiSpot collection kit, which includes:

  • Step-by-step instructions for collecting a blood sample.
  • Secure packaging for sample protection.
  • Details on how to return your sample to our laboratory.

Once received, your sample is processed under strict internal quality controls.
Results are typically available within 14 business days from the date the sample arrives at our facility. All analyses are performed using validated molecular and biochemical techniques in Biocentaur’s certified laboratories, ensuring the highest standards of accuracy and reproducibility.
