How does the PrimeSpot test work?

PrimeSpot test can accurately diagnose infections from different viruses, Lyme disease and chronic fatigue syndrome using sensitive molecular biology techniques (quantitative Polymerase Chain Reaction – qPCR). During the PrimeSpot test, a patient’s DNA and RNA is isolated from blood cells and serum. We then use them as a template in both one-step and two-step RNA qPCR reactions. The PrimeSpot test is highly sensitive and can detect and quantify certain viruses, and the presence of Lyme disease-associated species.

What is the scientific basis for the PrimeSpot test?

Quantitative Polymerase chain reaction (qPCR) techniques have been performed using primers for viruses HHVs, VZV, CMV, EBV, HTLV1, HPV16, HPV 18 and XMVR. A direct PCR has been performed for DNA viruses. A two-step qPCR has been performed with relative quantification for retroviruses. The measuring of cytokines has been performed using gene expression profile using inflammatory markers.

How are PrimeSpot samples analysed?

White cells isolated and serum is collected from blood sample. DNA or RNA (retroviruses) is then isolated and used as template in one-step or two-step RNA (retroviruses) qPCR reactions, using appropriate positive and negative controls. The gene expression profile of inflammatory markers is performed in mRNA isolated from specific cell-population. The above cells have been selected and isolated using antibodies conjugated with magnetic beads or fluorescence activated cell sorting. The final report includes information about the presence of each strain identified, as well as a quantification parameter (Ct, or threshold cycle).

How accurate is the PrimeSpot test?

All reactions are performed in triplicate. In addition positive and appropriate negative controls ensure accurate and reliable results.
